Cannot save changes to "Web.config" file - it says access is denied

Cannot save changes to "Web.config" file - it says access is denied
I am getting an "Internal Error" when I makes changes to shipping configuration.  I tried to turn off the custom error messaging in "Web.config" so that I can see what the problem is, but when I made the appropriate change to "Off" and tried to save the change, it wont let me and tells me access is denied.?!   I am signed in as Administrator and I double checked the permissions on the Web.config file as well as the "Web" directory and both allow administrator FULL control. 
 
I need help asap to resolve this since it is affecting my shipping options that I cannot change.

Check if it has read-only attribute on or restart web server if the file is locked up. Anyway you can update the exception notification email to yourself in web.config so that you will receive the email when there is an "internal error".
